    This deliverable describes the organisation, results and validation phase of the second Gender STI Co-Design Lab (hereafter the Lab) involving participants from America and Europe. The Co-design Lab that took place between October and November 2022 addressed the Gender STI objectives to integrate the gender perspective in bilateral and multilateral agreements between the EU Member States (MS), Associated Countries (AC) and third countries through design thinking methods and participatory techniques. The document describes the methods, participatory steps and tools that have been applied in the Lab to co-design shared solutions and prototypes for common challenges regarding gender inequalities in STI and to support the emergence of an international community of practitioners with similar challenging objectives. More specifically, the deliverable collects in detail “what” was done and how it was done in the Lab, reflects on the “so what” question, as to the sense and purpose of the challenge-based prototypes and their initial outputs, and finally draws some conclusions with a “now what” reflection on what was learnt and suggests possible priorities for future actions.

    Background: We wished to compare the nuisance parameters of pediatric vs. adult randomized-trials (RCTs) and determine if the latter can be used in sample size computations of the former.Methods: In this meta-epidemiologic empirical evaluation we examined meta-analyses from the Cochrane Database of Systematic-Reviews, with at least one pediatric-RCT and at least one adult-RCT. Within each meta-analysis of binary efficacy-outcomes, we calculated the pooled-control-group event-rate (CER) across separately all pediatric and adult-trials, using random-effect models and subsequently calculated the control-group event-rate risk-ratio (CER-RR) of the pooled-pediatric-CERs vs. adult-CERs. Within each meta-analysis with continuous outcomes we calculated the pooled-control-group effect standard deviation (CE-SD) across separately all pediatric and adult-trials and subsequently calculated the CE-SD-ratio of the pooled-pediatric-CE-SDs vs. adult-CE-SDs. We then calculated across all meta-analyses the pooled-CER-RRs and pooled-CE-SD-ratios (primary endpoints) and the pooled-magnitude of effect-sizes of CER-RRs and CE-SD-ratios using REMs. A ratio < 1 indicates that pediatric trials have smaller nuisance parameters than adult trials.Results: We analyzed 208 meta-analyses (135 for binary-outcomes, 73 for continuous-outcomes). For binary outcomes, pediatric-RCTs had on average 10% smaller CERs than adult-RCTs (summary-CE-RR: 0.90; 95% CI: 0.83, 0.98). For mortality outcomes the summary-CE-RR was 0.48 (95% CIs: 0.31, 0.74). For continuous outcomes, pediatric-RCTs had on average 26% smaller CE-SDs than adult-RCTs (summary-CE-SD-ratio: 0.74).Conclusions: Clinically relevant differences in nuisance parameters between pediatric and adult trials were detected. These differences have implications for design of future studies. Extrapolation of nuisance parameters for sample-sizes calculations from adult-trials to pediatric-trials should be cautiously done.

    During an inventory of insect diversity on Anticosti Island in 1993, we caught unprecedented numbers of Neospondylisupiformis (Mannerheim), a longhorned beetle rarely observed in eastern North America. All specimens were caught using 12-funnel Lindgren traps baited with 95% ethanol and α-pinene. This longhorned beetle was captured again in 2007 on Anticosti with the same traps. Other than that, seven specimens of N.upiformis were caught elsewhere in Quebec between 1993 and 2015. Only 14 specimens were found in the 45 most important insect collections of the province, the most recent specimen dating back to 1964. At least 90% of the captures came from old-growth balsam fir stands of the south-central part of the island. Seasonal flight activity ranged from early June to late July, but adult captures peaked in early July. Results suggest that Anticosti Island might be a hot spot for N.upiformis in eastern North America, particularly in its south-central part where old-growth balsam fir forests still exist.

    This study assessed the impact of 8 weeks biweekly in-season weightlifting training on the strength, throwing ability, and body composition of healthy male handball players. Twenty players (age: 21.2 ± 0.7 years, height: 1.83 ± 0.08 m, body mass: 83.3 ± 7.5 kg, body fat: 13.2 ± 1.4%, upper limb muscle volume: 3.16 ± 0.16 L) were randomly allocated between experimental (EG) and control (CG) groups. Measures of one-repetition maximal strength included bench press, pull-over, snatch, and clean and jerk. Throwing velocity was investigated by standing, running, and jump throws, and the power of the upper limbs was estimated from the total distance of a 3-kg medicine ball overhead throw. Muscle volumes were estimated anthropometrically. Training sessions comprised 3−4 sets of explosive weightlifting exercise at 75%−90% of 1RM (repetition maximum). Significant interaction effects (time x group) were found for all strength and throwing variables, ranging from ηp2 = 0.595 (pull-over) to ηp2 = 0.887 (medicine ball throw), with the largest between-group difference (more than 40%, Δd = 6.65) and effect size (d = 6.44) for the medicine ball throw, and the smallest (about 23%, Δd = 1.61) for the standing shot performance. Significant interaction effects were also detected for all anthropometric parameters (body mass: ηp2 = 0.433; body fat: ηp2 = 0.391; upper limb muscle volume: ηp2 = 0.920, with an almost 20% gain of muscle volume). It can be concluded that 8 weeks of biweekly in-season weightlifting training yielded substantial increases of muscle volume, maximal strength of the upper limbs, and ball throwing velocity in healthy handball players relative to their standard training program.

    N-methylation is a recurring feature in the biosynthesis of many plant specialized metabolites, including alkaloids. A crucial step in the conserved central pathway that provides intermediates for the biosynthesis of benzylisoquinoline alkaloids (BIAs) involves conversion of the secondary amine (S)-coclaurine into the tertiary amine (S)-N-methylcoclaurine by coclaurine N-methyltransferase (CNMT). Subsequent enzymatic steps yield the core intermediate (S)-reticuline, from which various branch pathways for the biosynthesis of major BIAs such as morphine, noscapine and sanguinarine diverge. An additional N-methylation yielding quaternary BIAs is catalyzed by reticuline N-methyltransferase (RNMT), such as in the branch pathway leading to the taxonomically widespread and ecologically significant alkaloid magnoflorine. Despite their functional differences, analysis of primary sequence information has been unable to accurately distinguish between CNMT-like and RNMT-like enzymes, necessitating laborious in vitro screening. Furthermore, despite a recent emphasis on structural characterization of BIA NMTs, the features and mechanisms underlying the CNMT-RNMT functional dichotomy were unknown. We report the identification of structural variants tightly correlated with function in known BIA NMTs and show through reciprocal mutagenesis that a single residue acts as a switch between CNMT- and RNMT-like functions. We use yeast in vivo screening to show that this discovery allows for accurate prediction of activity strictly from primary sequence information and, on this basis, improve the annotation of previously reported putative BIA NMTs. Our results highlight the unusually short mutational distance separating ancestral CNMT-like enzymes from more evolutionarily advanced RNMT-like enzymes, and thus help explain the widespread yet sporadic occurrence of quaternary BIAs in plants. While this is the first report of structural variants controlling mono-versus di-methylation activity among plant NMT enzymes, comparison with bacterial MT enzymes also suggests possible convergent evolution.

    The Niphta Theischinger fauna of South America is revised to include 11 species, nine of which are described as new to science (N. acus Pivar, sp. nov., N. bifurcata Pivar &amp; Moulton, sp. nov., N. bispinosa Pivar &amp; Sinclair, sp. nov., N. brunnea Pivar, sp. nov., N. courtneyi Pivar, sp. nov., N. daniellae Pivar, sp. nov., N. downesi Pivar, sp. nov., N. eurydactyla Pivar, sp. nov., N. mapuche Pivar, sp. nov.). The genus Niphta is redefined, both previously described Chilean species are redescribed, N. halteralis (Edwards) and N. nudipennis (Edwards), and females are described or redescribed where possible. The first descriptions of the immature stages of South American Niphta are provided, which represent a new larval morphotype in Thaumaleidae, as larvae and pupae possess ventral adhesive structures. Furthermore, these larvae were collected from vegetation rather than rocky substrates. Illustrations and micrographs are provided for all species, and scanning electron microscopy images are included for select immatures. A key to species, distribution maps, and discussions regarding phylogenetic affinities and habitat are also included.

    Background: High occupational stress has been associated with altered eating behaviors and obesity. Occupational stress is reported to be high in Asian countries. Furthermore, many Asian countries are increasingly consuming Western-type foods (e.g., incorporating drinks with meals) which collectively may also be contributing to obesity. Therefore, the aim of this study was to examine (a) associations between sugar-sweetened beverage (SSB) consumption as meal replacement and obesity and (b) associations between workload and substituting meals with SSB in nurses. Methods: A representative sample of 854 hospital-based nurses completed a structured questionnaire about SSB consumption, workload, and body mass index (BMI). Log binomial regression models were employed to test associations between SSBs and obesity rates and associations between workload and SSBs. Results: Most participants (57.6%) consumed SSBs as meal replacements during work. This was related to high workloads during shifts. Substituting SSBs for meals was significantly associated with increased likelihood of obesity (aPRR = 1.4, 95% CI (1.1, 1.7)). Workload was positively associated with SSB intake as meal substitutes (aPRR = 1.4, 95% CI (1.2, 1.6)). Conclusions: Our findings show that SSBs are used as meal substitutes and is due to the workload demands. Sugar-sweetened beverage consumption is also positively associated with the increased likelihood of obesity. Interventions that modify workloads and decrease SSB consumption may improve workers’ eating behaviors and health.

    Dam building and reservoir operations alter the downstream hydrological regime, and as a result, affect the health of the river aquatic ecosystem, particularly for large-scale cascade reservoirs. This study investigated the impact of the Gezhouba Reservoir (GR) and the Three Gorges Reservoir (TGR) on the spawning conditions of two critical taxa, i.e., the endemic four major carps and the endangered Chinese sturgeon in the Yangtze River. We analyzed the flow, sediment, and thermal regime in these two taxa spawning seasons and compared their features between the predam and postdam periods. Our results revealed that the GR and the TGR had altered the frequency distributions of flow, sediment, and water temperature to different degrees, with the impact by the GR on the carps and Chinese sturgeon ranked as water temperature > flow, sediment > water temperature > flow, and the effect of the TGR on these two taxa were ordered as flow > water temperature, sediment > flow > water temperature. For the GR, the satisfying degree of the suitable flow and water temperature of the carps increased, whilst the suitable flow, sediment, and water temperature for the Chinese sturgeon decreased. These changes in TGR showed a significant ascending (descending) trend in the suitable flow (water temperature) for the carps, and a clear decreasing trend in the flow, sediment, and temperature for Chinese sturgeon. Both the TGR and the GR had negative impacts on the spawning of these two taxa in terms of the rising/falling flow characteristics.

    Education is a common strategy used to prevent occupational injuries. However, its effectiveness is often measured using surrogate measures instead of true injury outcomes. To evaluate the effectiveness of workplace educational interventions, we selectively analyzed studies that reported injury outcomes (PROSPERO ID: CRD42019140631). We searched databases for peer-reviewed journal articles and sources of grey literature such as abstracts, registered trials, and theses published between 2000 and 2019. Studies on educational interventions that reported fatal or non-fatal occupational injury outcomes were selected. Two reviewers independently and in duplicate screened the studies, extracted data, and assessed risk of bias. Heterogeneity in the data precluded meta-analysis, and the results were reviewed narratively. In total, 35 studies were included. Of which, 17 found a significant reduction in injuries, most of which featured a multifaceted approach or non-didactic education. The remaining studies either described equivocal results or did not report statistical significance. Overall, interventions in the manufacturing industry were more effective than those in the construction sector. Risk of bias among included studies was moderate to high. In conclusion, educational interventions could be an effective part of multifaceted injury prevention programs. However, over-reliance on didactic education alone is not advised.

    Along with the rapid urbanization process in Beijing, China, the number of older rural-urban migrants is increasing. This study aims to understand how Chinese rural-urban migration in older age is influenced by, and impacts on the migrants’ mobilities. This study draws on a new conceptual framework of mobile vulnerability, influenced by physical, economic, institutional, social and cultural mobility, to understand older people’ experiences of migration from rural to urban areas. Forty-five structured in-depth interviews with older rural-urban migrants aged 55 and over were undertaken in four study sites in Beijing, using the constant comparative method. Results demonstrate that rural household registration (hukou) is an important factor that restricts rural older migrants’ institutional mobility. As older migrants’ physical mobility declines, their mobile vulnerability increases. Economic mobility is the key factor that influences their intention to stay in Beijing. Older migrants also described coping strategies to improve their socio-cultural mobility post-migration. These findings will inform service planning for older rural-urban migrants aimed at maintaining their health and wellbeing.

Furthermore, these larvae were collected from vegetation rather than rocky substrates. Illustrations and micrographs are provided for all species, and scanning electron microscopy images are included for select immatures. A key to species, distribution maps, and discussions regarding phylogenetic affinities and habitat are also included.

    Background: High occupational stress has been associated with altered eating behaviors and obesity. Occupational stress is reported to be high in Asian countries. Furthermore, many Asian countries are increasingly consuming Western-type foods (e.g., incorporating drinks with meals) which collectively may also be contributing to obesity. Therefore, the aim of this study was to examine (a) associations between sugar-sweetened beverage (SSB) consumption as meal replacement and obesity and (b) associations between workload and substituting meals with SSB in nurses. Methods: A representative sample of 854 hospital-based nurses completed a structured questionnaire about SSB consumption, workload, and body mass index (BMI). Log binomial regression models were employed to test associations between SSBs and obesity rates and associations between workload and SSBs. Results: Most participants (57.6%) consumed SSBs as meal replacements during work. This was related to high workloads during shifts. Substituting SSBs for meals was significantly associated with increased likelihood of obesity (aPRR = 1.4, 95% CI (1.1, 1.7)). Workload was positively associated with SSB intake as meal substitutes (aPRR = 1.4, 95% CI (1.2, 1.6)). Conclusions: Our findings show that SSBs are used as meal substitutes and is due to the workload demands. Sugar-sweetened beverage consumption is also positively associated with the increased likelihood of obesity. Interventions that modify workloads and decrease SSB consumption may improve workers’ eating behaviors and health.

    Dam building and reservoir operations alter the downstream hydrological regime, and as a result, affect the health of the river aquatic ecosystem, particularly for large-scale cascade reservoirs. This study investigated the impact of the Gezhouba Reservoir (GR) and the Three Gorges Reservoir (TGR) on the spawning conditions of two critical taxa, i.e., the endemic four major carps and the endangered Chinese sturgeon in the Yangtze River. We analyzed the flow, sediment, and thermal regime in these two taxa spawning seasons and compared their features between the predam and postdam periods. Our results revealed that the GR and the TGR had altered the frequency distributions of flow, sediment, and water temperature to different degrees, with the impact by the GR on the carps and Chinese sturgeon ranked as water temperature > flow, sediment > water temperature > flow, and the effect of the TGR on these two taxa were ordered as flow > water temperature, sediment > flow > water temperature. For the GR, the satisfying degree of the suitable flow and water temperature of the carps increased, whilst the suitable flow, sediment, and water temperature for the Chinese sturgeon decreased. These changes in TGR showed a significant ascending (descending) trend in the suitable flow (water temperature) for the carps, and a clear decreasing trend in the flow, sediment, and temperature for Chinese sturgeon. Both the TGR and the GR had negative impacts on the spawning of these two taxa in terms of the rising/falling flow characteristics.

    Education is a common strategy used to prevent occupational injuries. However, its effectiveness is often measured using surrogate measures instead of true injury outcomes. To evaluate the effectiveness of workplace educational interventions, we selectively analyzed studies that reported injury outcomes (PROSPERO ID: CRD42019140631). We searched databases for peer-reviewed journal articles and sources of grey literature such as abstracts, registered trials, and theses published between 2000 and 2019. Studies on educational interventions that reported fatal or non-fatal occupational injury outcomes were selected. Two reviewers independently and in duplicate screened the studies, extracted data, and assessed risk of bias. Heterogeneity in the data precluded meta-analysis, and the results were reviewed narratively. In total, 35 studies were included. Of which, 17 found a significant reduction in injuries, most of which featured a multifaceted approach or non-didactic education. The remaining studies either described equivocal results or did not report statistical significance. Overall, interventions in the manufacturing industry were more effective than those in the construction sector. Risk of bias among included studies was moderate to high. In conclusion, educational interventions could be an effective part of multifaceted injury prevention programs. However, over-reliance on didactic education alone is not advised.

    Along with the rapid urbanization process in Beijing, China, the number of older rural-urban migrants is increasing. This study aims to understand how Chinese rural-urban migration in older age is influenced by, and impacts on the migrants’ mobilities. This study draws on a new conceptual framework of mobile vulnerability, influenced by physical, economic, institutional, social and cultural mobility, to understand older people’ experiences of migration from rural to urban areas. Forty-five structured in-depth interviews with older rural-urban migrants aged 55 and over were undertaken in four study sites in Beijing, using the constant comparative method. Results demonstrate that rural household registration (hukou) is an important factor that restricts rural older migrants’ institutional mobility. As older migrants’ physical mobility declines, their mobile vulnerability increases. Economic mobility is the key factor that influences their intention to stay in Beijing. Older migrants also described coping strategies to improve their socio-cultural mobility post-migration. These findings will inform service planning for older rural-urban migrants aimed at maintaining their health and wellbeing.
