    Virus-based expression systems have been widely exploited for the production of recombinant proteins in plants during the last three decades. Advances in technology have boosted scale-up manufacturing of plant-made pharmaceuticals to high levels, via the complementation of transient expression and viral vectors. This combination allows proteins of interest to be produced in plants within a matter of days and thus, is well suited for the development of plant-made vaccines or therapeutics against emerging infectious diseases and potential bioterrorism agents. Several plant-based products are currently in varying stages of clinical development. To investigate the viability of virus-based expression systems for plant-made subunit vaccines against Porcine reproductive and respiratory syndrome virus (PRRSV), I have developed several viral vectors which express partial PRRSV glycoprotein 5 (GP5). In this thesis, I demonstrated that viral vectors can increase the expression of proteins for vaccine development in a variety of host plants. Green fluorescent protein (GFP) and the C-terminus of GP5 fused with GFP (GP5C-GFP) were expressed at up to 37.29 mg/kg and 4.36 mg/kg fresh weight of leaf tissue, respectively, in soybean plants via biolistic bombardment, using viral vectors derived from Bean pod mottle virus (BPMV). A Cucumber green mottle mosaic virus (CGMMV)-based vector was employed to display chimeric virus particles, presenting the neutralizing epitope (NE) of GP5, at levels as high as 35.84 mg/kg of cucumber leaf fresh weight, via agroinfiltration. In addition, GFP and the ectodomain of GP5 fused with GFP (GP5e-GFP) were successfully produced at up to 2.03 g/kg and 36.53 mg/kg fresh weight of leaf tissue, respectively, in Nicotiana benthamiana using transient expression of Pepino mosaic virus (PepMV)-based vectors. Co-infiltration of viral vectors with p19, a silencing suppressor, can help to mitigate the toxicity of GP5e to plant tissue and enhance the accumulation of this protein. These plant-made products may be tested as oral subunit vaccines against PRRSV in pigs during future experimental trials. Overall, the results demonstrate that viral systems can produce low-cost, versatile and robust vaccines that have a great impact in the fight against viral diseases, especially in developing countries.

    In this paper, we extend recent results of adaptive extremum seeking control to a class of nonlinear distributed parameter system. We address the real-time optimization of a chemical reaction that occurs in a tubular reactor described by a hyperbolic set of partial differential equations. An estimation and a control algorithm that take into account temperature constraints are developed based on a Lyapunov functional. The result of this algorithm is a late-lumping feedback profile control that steers the system to its optimum.

    Implicit solvent models offer an attractive way to estimate the effects of a solvent environment on the properties of small or large solutes without the complications of explicit simulations. One common test of accuracy is to compute the free energy of transfer from gas to liquid for a variety of small molecules, since many of these values have been measured. Studies of the temperature dependence of these values (i.e. solvation enthalpies and entropies) can provide additional insights into the performance of implicit solvent models. Here, we show how to compute temperature derivatives of hydration free energies for the 3D-RISM integral equation approach. We have computed hydration free energies of 1123 small drug-like molecules (both neutral and charged). Temperature derivatives were also used to calculate hydration energies and entropies of 74 of these molecules (both neutral and charged) for which experimental data is available. While direct results have rather poor agreement with experiment, we have found that several previously proposed linear hydration free energy correction schemes give good agreement with experiment. These corrections also provide good agreement for hydration energies and entropies though simple extensions are required in some cases.

    This paper extends the notion of the $\la$-coalescent of Pitman (1999) to the spatial setting. The partition elements of the spatial $\Lambda$-coalescent migrate in a (finite) geographical space and may only coalesce if located at the same site of the space. We characterize the $\Lambda$-coalescents that come down from infinity, in an analogous way to Schweinsberg (2000). Surprisingly, all spatial coalescents that come down from infinity, also come down from infinity in a uniform way. This enables us to study space-time asymptotics of spatial $\Lambda$-coalescents on large tori in $d\ge 3$ dimensions. Our results generalize and strengthen those of Greven et al. (2005), who studied the spatial Kingman coalescent in this context. Comment: 29 pages

    Cet article étudie le lien entre l’efficacité de l’intermédiation financière et la croissance économique sur un panel de sept pays de l’UEMOA** sur la période 1990-2008. Tout d’abord, un modèle théorique est présenté dans lequel l’efficacité de l’intermédiation bancaire renforce la croissance économique par le biais de l’amélioration de la productivité marginale du capital. Ensuite, l’efficacité du secteur bancaire est étudiée par le concept d’efficacité-X de Leibenstein (1966). Les scores d’efficacité bancaire sont estimés par la méthode DEA (Data Envelopment Analysis), appliquée à un modèle de production bancaire sur un échantillon de banques de l’UEMOA. Finalement, les scores moyens d’efficacité bancaire par pays sont calculés et utilisés comme indicateur de l’efficacité de l’intermédiation financière dans un modèle de croissance sur données de panel. Les résultats suggèrent l’existence d’une influence positive et significative de l’efficacité du secteur bancaire sur la croissance économique. This paper explores the connection between financial intermediation efficiency and economic growth on a panel of seven countries of WAEMU from 1990 to 2008. First, a theoretical model is presented in which financial efficiency enhances economic growth by means of increasing the marginal productivity of capital. Next, the banking sector efficiency is apprehended by the X-efficiency concept of Leibenstein (1966). Banking efficiency scores are estimated with Data Envelopment Analysis method, applied to banking production model on a sample of banks of the WAEMU. Finally, average scores of banking efficiency by country are calculated and used as indicator of financial intermediation efficiency in a growth model on panel data. Results suggest the existence of a positive impact of the banking sector efficiency on the economic growth.

L'histoire verticale de la philosophie peut se faire selon deux méthodes différentes : la méthode des sources et la méthode des structures. L'auteur montre la supériorité de la méthode des structures pour dégager l'intelligibilité interne de l'oeuvre, méthode qu'il présente comme une méthode des raisons. La méthode des structures correspond à la nature de l'oeuvre philosophique, d'une part, en tant qu'oeuvre, d'autre part, en tant que philosophique. The author shows the principles of the method that he applies in his studies on the history of philosophy. He distinguishes between the horizontal history of philosophy that interests primarily on the historical aspects of the philosophical systems and the vertical history of philosophy that interests at first to the philosophical doctrines. The vertical history of philosophy can be made according to two different methods : the method of source and the method of structures. The author shows the superiority of the method of structures to grasp the internal intelligibility of the philosophical work, method presented as a methodof reasons. The method of structures is adequate to the nature of the philosophical work, as work and as philosophical.

    Virus-based expression systems have been widely exploited for the production of recombinant proteins in plants during the last three decades. Advances in technology have boosted scale-up manufacturing of plant-made pharmaceuticals to high levels, via the complementation of transient expression and viral vectors. This combination allows proteins of interest to be produced in plants within a matter of days and thus, is well suited for the development of plant-made vaccines or therapeutics against emerging infectious diseases and potential bioterrorism agents. Several plant-based products are currently in varying stages of clinical development. To investigate the viability of virus-based expression systems for plant-made subunit vaccines against Porcine reproductive and respiratory syndrome virus (PRRSV), I have developed several viral vectors which express partial PRRSV glycoprotein 5 (GP5). In this thesis, I demonstrated that viral vectors can increase the expression of proteins for vaccine development in a variety of host plants. Green fluorescent protein (GFP) and the C-terminus of GP5 fused with GFP (GP5C-GFP) were expressed at up to 37.29 mg/kg and 4.36 mg/kg fresh weight of leaf tissue, respectively, in soybean plants via biolistic bombardment, using viral vectors derived from Bean pod mottle virus (BPMV). A Cucumber green mottle mosaic virus (CGMMV)-based vector was employed to display chimeric virus particles, presenting the neutralizing epitope (NE) of GP5, at levels as high as 35.84 mg/kg of cucumber leaf fresh weight, via agroinfiltration. In addition, GFP and the ectodomain of GP5 fused with GFP (GP5e-GFP) were successfully produced at up to 2.03 g/kg and 36.53 mg/kg fresh weight of leaf tissue, respectively, in Nicotiana benthamiana using transient expression of Pepino mosaic virus (PepMV)-based vectors. Co-infiltration of viral vectors with p19, a silencing suppressor, can help to mitigate the toxicity of GP5e to plant tissue and enhance the accumulation of this protein. These plant-made products may be tested as oral subunit vaccines against PRRSV in pigs during future experimental trials. Overall, the results demonstrate that viral systems can produce low-cost, versatile and robust vaccines that have a great impact in the fight against viral diseases, especially in developing countries.

    In this paper, we extend recent results of adaptive extremum seeking control to a class of nonlinear distributed parameter system. We address the real-time optimization of a chemical reaction that occurs in a tubular reactor described by a hyperbolic set of partial differential equations. An estimation and a control algorithm that take into account temperature constraints are developed based on a Lyapunov functional. The result of this algorithm is a late-lumping feedback profile control that steers the system to its optimum.

    Implicit solvent models offer an attractive way to estimate the effects of a solvent environment on the properties of small or large solutes without the complications of explicit simulations. One common test of accuracy is to compute the free energy of transfer from gas to liquid for a variety of small molecules, since many of these values have been measured. Studies of the temperature dependence of these values (i.e. solvation enthalpies and entropies) can provide additional insights into the performance of implicit solvent models. Here, we show how to compute temperature derivatives of hydration free energies for the 3D-RISM integral equation approach. We have computed hydration free energies of 1123 small drug-like molecules (both neutral and charged). Temperature derivatives were also used to calculate hydration energies and entropies of 74 of these molecules (both neutral and charged) for which experimental data is available. While direct results have rather poor agreement with experiment, we have found that several previously proposed linear hydration free energy correction schemes give good agreement with experiment. These corrections also provide good agreement for hydration energies and entropies though simple extensions are required in some cases.

    This paper extends the notion of the $\la$-coalescent of Pitman (1999) to the spatial setting. The partition elements of the spatial $\Lambda$-coalescent migrate in a (finite) geographical space and may only coalesce if located at the same site of the space. We characterize the $\Lambda$-coalescents that come down from infinity, in an analogous way to Schweinsberg (2000). Surprisingly, all spatial coalescents that come down from infinity, also come down from infinity in a uniform way. This enables us to study space-time asymptotics of spatial $\Lambda$-coalescents on large tori in $d\ge 3$ dimensions. Our results generalize and strengthen those of Greven et al. (2005), who studied the spatial Kingman coalescent in this context. Comment: 29 pages
