    Virus-based expression systems have been widely exploited for the production of recombinant proteins in plants during the last three decades. Advances in technology have boosted scale-up manufacturing of plant-made pharmaceuticals to high levels, via the complementation of transient expression and viral vectors. This combination allows proteins of interest to be produced in plants within a matter of days and thus, is well suited for the development of plant-made vaccines or therapeutics against emerging infectious diseases and potential bioterrorism agents. Several plant-based products are currently in varying stages of clinical development. To investigate the viability of virus-based expression systems for plant-made subunit vaccines against Porcine reproductive and respiratory syndrome virus (PRRSV), I have developed several viral vectors which express partial PRRSV glycoprotein 5 (GP5). In this thesis, I demonstrated that viral vectors can increase the expression of proteins for vaccine development in a variety of host plants. Green fluorescent protein (GFP) and the C-terminus of GP5 fused with GFP (GP5C-GFP) were expressed at up to 37.29 mg/kg and 4.36 mg/kg fresh weight of leaf tissue, respectively, in soybean plants via biolistic bombardment, using viral vectors derived from Bean pod mottle virus (BPMV). A Cucumber green mottle mosaic virus (CGMMV)-based vector was employed to display chimeric virus particles, presenting the neutralizing epitope (NE) of GP5, at levels as high as 35.84 mg/kg of cucumber leaf fresh weight, via agroinfiltration. In addition, GFP and the ectodomain of GP5 fused with GFP (GP5e-GFP) were successfully produced at up to 2.03 g/kg and 36.53 mg/kg fresh weight of leaf tissue, respectively, in Nicotiana benthamiana using transient expression of Pepino mosaic virus (PepMV)-based vectors. Co-infiltration of viral vectors with p19, a silencing suppressor, can help to mitigate the toxicity of GP5e to plant tissue and enhance the accumulation of this protein. These plant-made products may be tested as oral subunit vaccines against PRRSV in pigs during future experimental trials. Overall, the results demonstrate that viral systems can produce low-cost, versatile and robust vaccines that have a great impact in the fight against viral diseases, especially in developing countries.

    Implicit solvent models offer an attractive way to estimate the effects of a solvent environment on the properties of small or large solutes without the complications of explicit simulations. One common test of accuracy is to compute the free energy of transfer from gas to liquid for a variety of small molecules, since many of these values have been measured. Studies of the temperature dependence of these values (i.e. solvation enthalpies and entropies) can provide additional insights into the performance of implicit solvent models. Here, we show how to compute temperature derivatives of hydration free energies for the 3D-RISM integral equation approach. We have computed hydration free energies of 1123 small drug-like molecules (both neutral and charged). Temperature derivatives were also used to calculate hydration energies and entropies of 74 of these molecules (both neutral and charged) for which experimental data is available. While direct results have rather poor agreement with experiment, we have found that several previously proposed linear hydration free energy correction schemes give good agreement with experiment. These corrections also provide good agreement for hydration energies and entropies though simple extensions are required in some cases.

    Cet article étudie le lien entre l’efficacité de l’intermédiation financière et la croissance économique sur un panel de sept pays de l’UEMOA** sur la période 1990-2008. Tout d’abord, un modèle théorique est présenté dans lequel l’efficacité de l’intermédiation bancaire renforce la croissance économique par le biais de l’amélioration de la productivité marginale du capital. Ensuite, l’efficacité du secteur bancaire est étudiée par le concept d’efficacité-X de Leibenstein (1966). Les scores d’efficacité bancaire sont estimés par la méthode DEA (Data Envelopment Analysis), appliquée à un modèle de production bancaire sur un échantillon de banques de l’UEMOA. Finalement, les scores moyens d’efficacité bancaire par pays sont calculés et utilisés comme indicateur de l’efficacité de l’intermédiation financière dans un modèle de croissance sur données de panel. Les résultats suggèrent l’existence d’une influence positive et significative de l’efficacité du secteur bancaire sur la croissance économique. This paper explores the connection between financial intermediation efficiency and economic growth on a panel of seven countries of WAEMU from 1990 to 2008. First, a theoretical model is presented in which financial efficiency enhances economic growth by means of increasing the marginal productivity of capital. Next, the banking sector efficiency is apprehended by the X-efficiency concept of Leibenstein (1966). Banking efficiency scores are estimated with Data Envelopment Analysis method, applied to banking production model on a sample of banks of the WAEMU. Finally, average scores of banking efficiency by country are calculated and used as indicator of financial intermediation efficiency in a growth model on panel data. Results suggest the existence of a positive impact of the banking sector efficiency on the economic growth.

    OBJECTIVE To describe the clinical epidemiology of extubation failure in a multicenter cohort of patients treated in pediatric cardiac ICUs. DESIGN Retrospective cohort study using prospectively collected clinical registry data. SETTING Pediatric Cardiac Critical Care Consortium registry. PATIENTS All patients admitted to the CICU at Pediatric Cardiac Critical Care Consortium hospitals. INTERVENTIONS None. MEASUREMENTS AND MAIN RESULTS Analysis of all mechanical ventilation episodes in the registry from October 1, 2013, to July 31, 2014. The primary outcome of extubation failure was reintubation less than 48 hours after planned extubation. Repeated-measures analysis using generalized estimating equations to account for within patient and center correlation was performed to identify risk factors for extubation failure. Adjusted extubation failure rates for each hospital were calculated using logistic regression controlling for patient factors. Of 1,734 mechanical ventilation episodes (1,478 patients at eight hospitals) ending in a planned extubation, there were 100 extubation failures (5.8%). In multivariable analysis, only longer duration of mechanical ventilation was significantly associated with extubation failure (p = 0.01); the failure rate was 4% when ventilated less than 24 hours, 9% after 24 hours, and 13% after 7 days. For 503 patients intubated and extubated in the cardiac operating room, 15 patients (3%) failed extubation within 48 hours (12 within 24 hr). Case-mix-adjusted extubation failure rates ranged from 1.1% to 9.8% across hospitals. Patients failing extubation had greater median cardiac ICU length of stay (15 vs 3 d; p < 0.001) and in-hospital mortality (7.9 vs 1.2%; p < 0.001). CONCLUSIONS Though extubation failure is uncommon overall, there may be opportunities to improve extubation readiness assessment in patients ventilated more than 24 hours. These data suggest that extubation in the operating room after cardiac surgery can be done with a low failure rate. We observed variation in extubation failure rates across hospitals, and future investigation must elucidate the optimal strategies of high-performing centers to reduce ventilation time while limiting extubation failures.

M’attachant plus particulièrement au travail de Dr Hill au sein du Comité pour l’adoption d’enfants de couleur et auprès de « Ontario Black History Society », j’avance que Dr Hill se définit comme étant « l’homme de la race noire », un terme masculiniste attribué à ceux qui cherchaient à diriger une communauté noire en Amérique du Nord et à effacer l’image infantilisée et féminisée du « mulâtre tragique », prisonnier dans un « monde gris pâle ». This paper is about Dr. Daniel Hill III, the first director of the Ontario Human Rights Agency. Paying particular attention to Dr Hill’s work with the Committee for the Adoption of Coloured Youngsters and the Ontario Black History Society, I argue that he fashioned himself as “Negro race man”, a masculinist term assigned to people who sought to lead a Black community in North America and lay to rest the infantilised and feminized image of the “tragic mulatto” trapped in a “light grey world”.

    In this paper, we present a general review of the status of numerical modelling applied to the design of high temperature superconductor (HTS) devices. The importance of this tool is emphasized at the beginning of the paper, followed by formal definitions of the notions of models, numerical methods and numerical models. The state-of-the-art models are listed, and the main limitations of existing numerical models are reported. Those limitations are shown to concern two aspects: one the one hand, the numerical performance (i.e. speed) of the methods themselves is not good enough yet; on the other hand, the availability of model file templates, material data and benchmark problems is clearly insufficient. Paths for improving those elements are provided in the paper. Besides the technical aspects of the research to be further pursued, for instance in adaptive numerical methods, most recommendations command for an increased collective effort for sharing files, data, codes and their documentation.

    This paper employs a descriptive case study method to analyze and critically review the emergence of the provincial poverty reduction strategy in Ontario, Canada which was implemented in 2008 and renewed in 2014. The purpose of this study is two-fold: first, it defines the principles of neoliberalism and explores the historical growth of neoliberal thought in Canada, and specifically within Ontario, beginning in the 1980s to the present-day. Drawing on a combination of primary, secondary and grey literature, this paper discusses the ways in which neoliberal ideologies and rhetoric became deeply rooted in political thought and discourse within the province. Employing a critical theory framework, the paper highlights the contrasting ways in which neoliberal values were adopted by the different political parties in power and the detrimental impact this espousal had on individuals living in poverty within Ontario. Second, the paper illustrates the powerful ways in which anti-poverty grassroots movements and social advocacy groups assembled to push for the creation of a provincial poverty reduction strategy. The analysis ends with a critique of the neoliberal influences on the strategy’s recommendations and the future outlook of the poverty reduction strategy based on the current political climate within the province.

    As energy systems grow more complex, modeling efforts spanning multiple scales, disciplines and perspectives are essential. Improved methods are needed to guide the development of not just individual models, but also multi-model ecologies - systems of interacting models. Currently there is a lack of knowledge concerning how multi-model ecologies can and should be designed to facilitate adequate understanding of energy system complexity and its consequences. Via an analysis of twelve multi-model initiatives both within and outside the energy domain, this paper elucidates possible design patterns and development paths for multi-model ecologies.The results highlight two broad paths to developing energy system multi-model ecologies, one prioritizing interoperability and the other prioritizing diversity. The former path facilitates the efficient development of models spanning multiple scales and (to a degree) disciplines, and can ease systematic testing of assumptions. The latter is suited to bridging traditional disciplines and perspectives and advancing knowledge within the interstices of different knowledge communities. It is furthermore suggested that a combination of diversity, connectivity and hierarchy in multi-model ecology composition is central to enabling the development of complex webs of models capable of addressing the complexity of real-world energy systems.

    Virus-based expression systems have been widely exploited for the production of recombinant proteins in plants during the last three decades. Advances in technology have boosted scale-up manufacturing of plant-made pharmaceuticals to high levels, via the complementation of transient expression and viral vectors. This combination allows proteins of interest to be produced in plants within a matter of days and thus, is well suited for the development of plant-made vaccines or therapeutics against emerging infectious diseases and potential bioterrorism agents. Several plant-based products are currently in varying stages of clinical development. To investigate the viability of virus-based expression systems for plant-made subunit vaccines against Porcine reproductive and respiratory syndrome virus (PRRSV), I have developed several viral vectors which express partial PRRSV glycoprotein 5 (GP5). In this thesis, I demonstrated that viral vectors can increase the expression of proteins for vaccine development in a variety of host plants. Green fluorescent protein (GFP) and the C-terminus of GP5 fused with GFP (GP5C-GFP) were expressed at up to 37.29 mg/kg and 4.36 mg/kg fresh weight of leaf tissue, respectively, in soybean plants via biolistic bombardment, using viral vectors derived from Bean pod mottle virus (BPMV). A Cucumber green mottle mosaic virus (CGMMV)-based vector was employed to display chimeric virus particles, presenting the neutralizing epitope (NE) of GP5, at levels as high as 35.84 mg/kg of cucumber leaf fresh weight, via agroinfiltration. In addition, GFP and the ectodomain of GP5 fused with GFP (GP5e-GFP) were successfully produced at up to 2.03 g/kg and 36.53 mg/kg fresh weight of leaf tissue, respectively, in Nicotiana benthamiana using transient expression of Pepino mosaic virus (PepMV)-based vectors. Co-infiltration of viral vectors with p19, a silencing suppressor, can help to mitigate the toxicity of GP5e to plant tissue and enhance the accumulation of this protein. These plant-made products may be tested as oral subunit vaccines against PRRSV in pigs during future experimental trials. Overall, the results demonstrate that viral systems can produce low-cost, versatile and robust vaccines that have a great impact in the fight against viral diseases, especially in developing countries.

    Implicit solvent models offer an attractive way to estimate the effects of a solvent environment on the properties of small or large solutes without the complications of explicit simulations. One common test of accuracy is to compute the free energy of transfer from gas to liquid for a variety of small molecules, since many of these values have been measured. Studies of the temperature dependence of these values (i.e. solvation enthalpies and entropies) can provide additional insights into the performance of implicit solvent models. Here, we show how to compute temperature derivatives of hydration free energies for the 3D-RISM integral equation approach. We have computed hydration free energies of 1123 small drug-like molecules (both neutral and charged). Temperature derivatives were also used to calculate hydration energies and entropies of 74 of these molecules (both neutral and charged) for which experimental data is available. While direct results have rather poor agreement with experiment, we have found that several previously proposed linear hydration free energy correction schemes give good agreement with experiment. These corrections also provide good agreement for hydration energies and entropies though simple extensions are required in some cases.

    Cet article étudie le lien entre l’efficacité de l’intermédiation financière et la croissance économique sur un panel de sept pays de l’UEMOA** sur la période 1990-2008. Tout d’abord, un modèle théorique est présenté dans lequel l’efficacité de l’intermédiation bancaire renforce la croissance économique par le biais de l’amélioration de la productivité marginale du capital. Ensuite, l’efficacité du secteur bancaire est étudiée par le concept d’efficacité-X de Leibenstein (1966). Les scores d’efficacité bancaire sont estimés par la méthode DEA (Data Envelopment Analysis), appliquée à un modèle de production bancaire sur un échantillon de banques de l’UEMOA. Finalement, les scores moyens d’efficacité bancaire par pays sont calculés et utilisés comme indicateur de l’efficacité de l’intermédiation financière dans un modèle de croissance sur données de panel. Les résultats suggèrent l’existence d’une influence positive et significative de l’efficacité du secteur bancaire sur la croissance économique. This paper explores the connection between financial intermediation efficiency and economic growth on a panel of seven countries of WAEMU from 1990 to 2008. First, a theoretical model is presented in which financial efficiency enhances economic growth by means of increasing the marginal productivity of capital. Next, the banking sector efficiency is apprehended by the X-efficiency concept of Leibenstein (1966). Banking efficiency scores are estimated with Data Envelopment Analysis method, applied to banking production model on a sample of banks of the WAEMU. Finally, average scores of banking efficiency by country are calculated and used as indicator of financial intermediation efficiency in a growth model on panel data. Results suggest the existence of a positive impact of the banking sector efficiency on the economic growth.
